چکیده

precise information about the interaction between retinylidene chromophore and protein moiety of (1) in the intermediates (1). In the previous paper (2), it had been chemically confirmed from the CD data (Table 1) of the rhodopsin analog (2) involving the 10-20 bonded structure that the ƒ¿-CD-band of (1) is due to the twisted 12s-trans-conformation in the chromophore. This is strong evidence supporting the theory for twisted chromophore (3-5) proposed for the induction of ƒ¿-CD-band in

Spectral sensitivity, structure and activation of eukaryotic rhodopsins: activation spectroscopy of rhodopsin analogs in Chlamydomonas.

Retinal normally binds opsin forming the chromophore of the visual pigment, rhodopsin. In this investigation synthetic analogs were bound by the opsin of living cells of the alga Chlamydomonas reinhardtii; the effect was assayed by phototaxis to give an activation spectrum for each rhodopsin analog. The results show the influence of different chromophores and the protein on the absorption of li...

Primary events in dim light vision: a chemical and spectroscopic approach toward understanding protein/chromophore interactions in rhodopsin.

The visual pigment rhodopsin (bovine) is a 40 kDa protein consisting of 348 amino acids, and is a prototypical member of the subfamily A of G protein-coupled receptors (GPCRs). This remarkably efficient light-activated protein (quantum yield = 0.67) binds the chromophore 11-cis-retinal covalently by attachment to Lys296 through a protonated Schiff base. The 11-cis geometry of the retinylidene c...

Mechanism of rhodopsin activation as examined with ring-constrained retinal analogs and the crystal structure of the ground state protein.

The guanine nucleotide-binding protein (G-protein)-coupled receptor superfamily (GPCR) is comprised of a large group of membrane proteins involved in a wide range of physiological signaling processes. The functional switch from a quiescent to an active conformation is at the heart of GPCR action. The GPCR rhodopsin has been studied extensively because of its key role in scotopic vision. The gro...

Chromophores in a Truly Heterogeneous Environment: Retinal and 13-demethyl-retinal inside the Rhodopsin Binding Pocket - a Qm/mm Study∗

To probe the effect of the protein environment on the retinal chromophore of rhodopsin, we have performed molecular dynamics simulations using combined quantum mechanics / molecular mechanics (QM/MM). The starting geometry of the protein is based on the 2.6Å X-ray structure of bovine rhodopsin by Okada et al. The wild type chromophore of rhodopsin shows a highly twisted conformation around the ...

Role of bulk water in hydrolysis of the rhodopsin chromophore.

Rhodopsin (Rho) is a prototypical G protein-coupled receptor that changes from an inactive conformational state to a G protein-activating state as a consequence of its retinal chromophore isomerization, 11-cis-retinal → all-trans-retinal. The photoisomerized chromophore covalently linked to Lys(296) by a Schiff base is subsequently hydrolyzed, but little is known about this reaction. Recent res...
